3. Ultrasonic parameter setting: Set the key and the working parameters of the instrument (see the manual or the laidian consultation for specific settings). For samples that are sensitive to temperature requirements (such as bacteria), an ice bath is generally used outside. The actual temperature must be lower than 25 degrees, and the protein Nucleic acids are definitely not denatured.

①   Time: The ultrasonic time should not exceed 5 seconds each time, and the interval time should be greater than or equal to the ultrasonic time to facilitate heat dissipation. The time setting should be based on the principle of short ultrasonic time and many ultrasonic times, which can prolong the life of the ultrasonic machine and the probe.

②  Ultrasonic power: It should not be too large to prevent the sample from splashing or foaming. If the sample volume is less than 10ml, the power should be within 200w, and a 2mm ultrasonic probe should be selected. In addition, set the horn selection switch on the back of the panel to the corresponding block; 10 - The power of the sample capacity of 200ml is 200-400w, use a 6mm ultrasonic probe, and set the horn behind the panel to the corresponding block; the power of the sample capacity above 200ml is between 300-600w, choose a 10mm ultrasonic probe, and place the panel The rear horn is hit to the corresponding block; (2MM small probe power is strictly prohibited to exceed 350W)

③   Vessel selection: choose as many beakers as there are samples, which is also conducive to the convection of samples in ultrasound and improves the crushing efficiency. For example; a 20ML beaker is best used for a 20ML processing capacity.
